To examine the effects of Dll1 blocking antibody (Dll1 Ab) on gene expression profiles of the brachiocephalic arteries, ApoE-deficient mice were fed a high-fat diet from 12 weeks of age. Dll1 Ab or control non-immune IgG was intraperitoneally injected from 20 weeks of age. At 32 weeks of age, brachiocephalic arteries were excised, and gene expressions were evaluated. To examine the effects of Dll1 Ab, 250 micrograms of the antibody (clone HMD1-5) were intraperitoneally injected twice a week. In the control group, an equivalent dose of non-immune IgG was administered similarly.
为探究Dll1阻断抗体(Dll1 blocking antibody,简称Dll1 Ab)对头臂动脉(brachiocephalic arteries)基因表达谱的影响,本实验以12周龄起接受高脂饮食喂养的载脂蛋白E缺陷型小鼠(ApoE-deficient mice)为研究对象。自20周龄起,向小鼠腹腔注射Dll1 Ab或对照非免疫免疫球蛋白G(non-immune IgG)。至32周龄时,摘取小鼠头臂动脉并对其基因表达水平进行评估。本实验中,实验组采用克隆号为HMD1-5的Dll1 Ab,剂量为250微克,每周腹腔注射两次;对照组则以相同给药方式给予等量的非免疫IgG。
