本研究采用染色质免疫共沉淀测序（ChIP-sequencing）技术，特异性分析MRN复合体（MRN Complex）与RNA聚合酶II（RNA polymerase II，RNAPII）在靶基因上的协同募集情况，并探究MRN复合体缺失对RNA聚合酶II及活跃转录过程的影响。
总体实验设计：将海拉细胞（HeLa）分别采用短发夹RNA（shRNA）、DRB处理，或进行热休克处理，随后对不同靶标因子开展染色质免疫共沉淀实验。
