RNAseq study of organs of Ixodes ricinus ticks harbouring or not the endosymbiont Midichloria mitochondrii

Compared transcriptomes of pools of organs (ovary, salivary glands or Malpighian tubules), from two strains of Ixodes ricinus, respectively with and without the symbiont Midichloria mitochondrii. Samples were obtained by pooling organs from n=5 adult female ticks. Two different engorgement time points (D0 i.e. unfed; D4 i.e. partially fed) were compared. RNA was extracted and the transcriptome was sequenced for each library, in order to identify differentially expressed genes between the different conditions.

本研究对两种分别携带与不携带共生菌Midichloria mitochondrii的蓖麻硬蜱（Ixodes ricinus）品系的器官混合样本（包括卵巢、唾液腺及马氏管）的转录组进行比较分析。样本通过混合n=5只成年雌性蜱虫的器官制备得到。本研究比较了两个不同的饱血时间点：D0即未吸血状态，D4即部分饱血状态。对每个测序文库提取RNA并进行转录组测序，以鉴定不同实验条件下的差异表达基因。
