本研究采用磷酸盐补料发酵策略,以延长色氨酸高产大肠杆菌(E. coli)菌株进入磷酸盐限制环境的过渡阶段。结合高频采样与该发酵策略,可获得磷酸盐响应相关调控机制的高分辨率解析结果。实验总体设计:以发酵2.5小时后的首个采样时间点作为参考时间点,该时间点对应磷酸盐充足条件。当磷酸盐浓度进入敏感区间后,根据发酵过程动态额外选取了9个采样时间点。
