An abdominal aortic aneurysm (AAA) is a pathological widening of the aortic wall characterized by loss of AoSMCs, extracellular matrix degradation and local inflammation. This condition is often asymptomatic until rupture occurs, leading to high morbidity and mortality rates. We conducted single-cell RNA sequencing (scRNA-seq) from AAA patients´s specimens to profile gene expression at single cell level and to gain insight on cell types relevant to disease dynamics. Overall design: To profile single-cell transcriptome and analyze the diversity of cell types present in human AAA tissue specimens, we carried out scRNA-seq from 4 AAA patients. Sequencing reactions were perfomed in duplicate.
腹主动脉瘤(abdominal aortic aneurysm, AAA)是一种主动脉壁的病理性扩张,其特征为主动脉平滑肌细胞(aortic smooth muscle cells, AoSMCs)丢失、细胞外基质降解以及局部炎症反应。该病症通常无明显临床症状,直至发生破裂,进而引发较高的发病率与死亡率。本研究对腹主动脉瘤患者的标本开展单细胞RNA测序(single-cell RNA sequencing, scRNA-seq),以实现单细胞水平的基因表达全景分析,并深入解析与疾病动态进程相关的细胞类型。总体实验设计:为对人类腹主动脉瘤组织标本的单细胞转录组进行全景分析并解析其中的细胞类型多样性,本研究对4名腹主动脉瘤患者的样本实施了单细胞RNA测序,且所有测序反应均以重复实验的方式开展。
