Kupffer cells (KCs) are the tissue resident macrophage population of the liver. With this experiment, we sought to investigate the potential consequences of decreasing embryonically-derived KC numbers on liver gene expression in the context of hypercholesterolemia. 3-month-old CD207-DTR x LDL-receptor deficient (C57BL6J background) female mice were injected with diphtheria toxin (DT) or heat-inactivated DT. Mice were then fed a cholesterol-rich diet to induce hypercholesterolemia. RNA sequencing was performed on liver RNA samples after 4 weeks of diet.
库普弗细胞(Kupffer cells, KCs)是肝脏的组织驻留巨噬细胞群体。本实验旨在探究高胆固醇血症背景下,胚胎起源的库普弗细胞数量减少对肝脏基因表达的潜在影响。将背景品系为C57BL/6J的3月龄CD207-DTR × 低密度脂蛋白受体缺陷雌性小鼠,分别注射白喉毒素(diphtheria toxin, DT)或热灭活白喉毒素。随后给小鼠喂食高胆固醇饲料以诱导高胆固醇血症,造模4周后对肝脏RNA样本开展RNA测序。
