T cells are key mediators of inflammation. Here we performed deep profiling of T cells in the gut of patients with inflammatory bowel disease to reveal the functional specialization of lymphocytes under pathological conditions and provide comprehensive insights into the pathogenesis of this disease. Overall design: Intestinal CD3+ T cells were isolated by Fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq. This technology was further combined to BD AbSeq system to measure protein and transcriptome levels simultaneously at a single cell level.

T细胞是炎症应答的关键介导因子。本研究对炎症性肠病患者肠道内的T细胞开展深度谱型分析,以揭示病理状态下淋巴细胞的功能特化,并为该疾病的发病机制提供全面的研究见解。 总体实验设计:通过荧光激活细胞分选(Fluorescence-activated cell sorting, FACS)分离肠道CD3+ T细胞,采用单细胞RNA测序(single-cell RNA sequencing, scRNAseq)进行分析;本研究进一步将该技术与BD AbSeq系统联用,可在单细胞层面同时检测蛋白质与转录组的表达水平。
