OsSKIPa is an SNW/SKIP protein in rice, which homolog to SKIP(human) and PRP45(yeast). OsSKIPa can rescue the lethal of yeast prp45 mutants. OsSKIPa-suppressed plants (S59R) show decrease of cell viability. Overexpression of OsSKIPa (S58S) increase the osmotic stresses tolerance. OsSKIPa is an important regulator and interacting with a lot of proteins which have varient functions. In order to find the genes expression information when OsSKIPa changed. We use microarray to find changed genes in S59R and S58S plants in the normal condition. Keywords: gene expression array-based (RNA / in situ oligonucleotide) Overall design: Whole plants were collected from 1-month-old S58S (two replicates), S59R (three replicates) and wild type (three replicates).
OsSKIPa是水稻中的SNW/SKIP家族蛋白,与人类SKIP及酵母PRP45具有同源性。OsSKIPa能够互补酵母prp45突变体的致死缺陷。OsSKIPa敲低植株(S59R)的细胞活力显著下降。过表达OsSKIPa的转基因植株(S58S)渗透胁迫耐受性显著增强。OsSKIPa是一类重要的调控因子,可与多种功能各异的蛋白质存在相互作用。为解析OsSKIPa表达改变后的基因表达谱特征,本研究通过微阵列(microarray)技术,在正常培养条件下对S58S、S59R及野生型植株中的差异表达基因进行筛选。关键词:基于基因表达芯片(RNA/原位寡核苷酸)。实验设计:采集生长1个月的S58S植株(2个生物学重复)、S59R植株(3个生物学重复)及野生型植株(3个生物学重复)的全株组织。
