RNA is more vulnerable to oxidation than other cellular components, which is linked to a range of diseases and pathological conditions, such as AKI. To identify the contribution of RNA oxidation to AKI, the mechanisms of how cells handle oxidized RNA should be elucidated. By RNA sequencing analysis in ischemia/reperfusion-induced AKI, we observed the significant changes of genes important in degradingting oxidative RNA. Overall design: The gene expression of 3 samples from a 24 h renal ischemia/reperfusion-induced AKI was compared to 3 sham samples by RNA sequencing.
RNA相较于其他细胞组分更易发生氧化损伤,这一现象与包括急性肾损伤(AKI)在内的多种疾病及病理状态存在紧密关联。为明确RNA氧化在急性肾损伤中的作用贡献,需先阐明细胞处理氧化RNA的具体机制。本研究通过对缺血再灌注诱导的急性肾损伤模型开展RNA测序分析,观察到降解氧化RNA的关键基因发生了显著表达变化。整体实验设计:通过RNA测序技术,本研究对比了3例经24小时肾缺血再灌注诱导的急性肾损伤样本与3例假手术样本的基因表达水平。
