Iron overload disorders are an important factor contributing to various pathologies. However, the effects of iron overload on liver transcriptional regulation are largely unknown. To examine the effects of iron overload on liver transcriptome and chromatin accessibility, we fed mice a high-iron diet (HID) for 2 weeks. Then, livers were collected and profiled by RNA-seq and ATAC-seq. 8 weeks old C57BL/6J male mice were fed chow (standard) diet (2018-Teklad Global 18% Protein Rodent Diet) and 2% carbonyl iron diet (TD 09521-2% carbonyl iron diet, Teklad) for 2 weeks (chronic treatment) and for 48 hours (acute treatment). Each group has 3 replicates.

铁过载紊乱是引发多种病变的重要诱因。然而,目前学界对铁过载调控肝脏转录过程的具体机制仍知之甚少。为探究铁过载对肝脏转录组(transcriptome)与染色质可及性(chromatin accessibility)的影响,本研究为小鼠饲喂高铁饮食(high-iron diet,HID),并设置2周的处理周期。随后采集小鼠肝脏组织,通过RNA-seq与ATAC-seq开展组学特征分析。实验选用8周龄的C57BL/6J雄性小鼠,分别饲喂普通标准饲料(2018-Teklad Global 18% Protein Rodent Diet)与2%羰基铁饲料(TD 09521-2% carbonyl iron diet, Teklad),处理时长分为2周(慢性处理)与48小时(急性处理),每组设置3个生物学重复。
