Myeloblast expansion is a hallmark of disease progression in leukemias and comprises of CD34+ hematopoeitic stem and progeneitor cells. We used single-cell RNA sequencing (scRNAseq) to analyze the transcriptional states of HSPCs in CMML. CD34+ HSPCs were isolated from bone marrow mononuclear cells from patients diagnosed with CMML and transcriptionally profiled using scRNAseq.

成髓细胞扩增是白血病疾病进展的标志性特征,其细胞群体由CD34+造血干祖细胞(hematopoietic stem and progenitor cells)构成。本研究借助单细胞RNA测序(single-cell RNA sequencing,scRNAseq)技术,对慢性粒单核细胞白血病(Chronic Myelomonocytic Leukemia,CMML)患者体内的造血干祖细胞(hematopoietic stem and progenitor cells,HSPCs)转录状态进行了分析。我们从确诊为CMML的患者骨髓单个核细胞中分离得到CD34+ HSPCs,并通过scRNAseq完成了其转录谱分析。
