Illustrating the cellular architecture of the human brain is critical to understanding its diverse functions and complex animal behaviors. Single nucleus methylation sequencing was applied to regions of human brains. we identified distinct cell clusters and formed them in a hierarchical taxonomy. These clusters include known primary brain cell types and possible sub-types. We use these data to identify the epigenomic characteristics and to define specific regulatory elements for each cell cluster. Apply snmC-seq3 to brain regions to identify epigenomic landcape of the adult human brains
阐明人脑的细胞架构,对于理解其多样功能与复杂动物行为至关重要。本研究对人脑多个脑区开展单细胞核甲基化测序(single nucleus methylation sequencing),鉴定得到多类独特的细胞簇,并构建层级分类学框架。这些细胞簇涵盖已报道的主要脑细胞类型及潜在亚型。本研究利用上述测序数据,解析各细胞簇的表观基因组特征,并明确其特异性调控元件。此外,本研究将snmC-seq3技术应用于脑区样本,以解析成人脑的表观基因组图谱(epigenomic landscape)。
