In order to indentify the transcriptional differences between GFI1 KD and GFI1 KO mice that may account for differences in occurence of AML between these mice, we performed exresiion profiling by RNA-Seq on healthy young and old mice mice from both genotypes. In the case of GFI1 KD, the older mice had developped AML whereas KO mice of comparable ages did not. Overall design: For each genotype (Gfi1 KD and GFI1 KO), RNA was extracted from the bone marrow of 2 young healthy mice as a biological duplicate. Bone marrow RNA was also extracted from 2 older GFI1 KD mice with AML and 2 healthy GFI1 KO mice of comparable ages

为了探究可解释两种小鼠急性髓系白血病(AML, Acute Myeloid Leukemia)发生差异的GFI1敲低(KD, Knockdown)与GFI1敲除(KO, Knockout)小鼠之间的转录组差异,我们对两种基因型的健康年轻与老年小鼠开展了RNA测序(RNA-Seq)表达谱分析。其中,GFI1 KD组的老年小鼠已罹患AML,而同年龄段的GFI1 KO组小鼠则未发生AML。整体实验设计:针对两种基因型(Gfi1 KD与GFI1 KO),我们从2只健康年轻小鼠的骨髓中提取RNA作为生物学重复样本;同时从2只已罹患AML的老年GFI1 KD小鼠,以及2只同年龄段健康GFI1 KO小鼠的骨髓中提取RNA。
