We here reported DNAJC12 levels are negatively correlated with the prognosis of children with neuroblastoma and the knockout of DNAJC12 improves cell proliferation and invasion in neuroblastoma cells.DNAJC12 knockout results in increase of intracellular lactic acid and H4K5la modification level. DNAJC12 affects COL1A1 gene transcription by regulating H4K5la modification level and plays an important role in neuroblastoma metastasis. 本研究报道,DNAJC12的表达水平与神经母细胞瘤患儿的预后呈负相关;敲除DNAJC12可提升神经母细胞瘤细胞的增殖与侵袭能力。DNAJC12敲除会导致细胞内乳酸水平及H4K5la修饰水平升高。DNAJC12可通过调控H4K5la修饰水平影响COL1A1基因的转录,在神经母细胞瘤的转移过程中发挥重要作用。为探究DNAJC12对SH-SY5Y细胞侵袭能力的调控功能,本研究通过CRISPR/Cas9技术构建了DNAJC12基因敲除的SH-SY5Y细胞系。
