We measued IgG autoantibodies associated with Connective Tissue Diseases (CTDs) and Anti-Cytokine Antibodies (ACA) in SLE and healthy controls who received the BNT162b2 vaccine. Overall design: We constructed protein arrays by first coupling recombinant protein antigens to uniquely fluorescent carboxylated magnetic beads. We then combined the beads into two seperate arrays that were used to probe patient SLE serum samples, healthy control samples, and autoimmune prototype samples. A Luminex FLEXMAP 3D® system was used to quantitatively measure autoantibody reactivity in each sample in units of mean flourescent intensity (MFI).

本研究检测了接种BNT162b2疫苗的系统性红斑狼疮(Systemic Lupus Erythematosus, SLE)患者与健康对照者体内与结缔组织病(Connective Tissue Diseases, CTDs)相关的IgG自身抗体以及抗细胞因子抗体(Anti-Cytokine Antibodies, ACA)。 整体实验设计:我们首先将重组蛋白抗原偶联至带有独特荧光标记的羧基化磁珠(carboxylated magnetic beads),以此构建蛋白芯片;随后将磁珠组合为两套独立的芯片,用于检测系统性红斑狼疮患者血清样本、健康对照血清样本以及自身免疫原型样本。本研究采用Luminex FLEXMAP 3D®系统,以平均荧光强度(mean fluorescent intensity, MFI)为单位,对每份样本中的自身抗体反应性进行定量检测。
