High-throughput measurements of gene expression as a function of gene position in Caulobacter crescentus

A barcoded operon was integrated into hundreds of positions on the chromosome using transposon mutagenesis. High-throughput sequencing was then used to extract both the barcoded mRNA and the genomic position of each integrated barcoded operon.一条带条形码的操纵子（barcoded operon）通过转座子诱变（transposon mutagenesis）技术被整合至染色体的数百个位点；随后借助高通量测序（high-throughput sequencing）技术，可同时提取得到带条形码的信使RNA（mRNA）以及每个已整合的带条形码操纵子的基因组插入位点。
