To study expression pattern of small nucleolar RNAs (snoRNAs) during influenza A viral infection, human cells A549 were infected with influenza A/Puerto Rico/8/1934 (H1N1) virus. Small RNA-seq analysis of infected cells after 24 h or 48 h incubation was performed on an Illumina NextSeq 500 platform. The same mock-infected cells were used as control. Small RNA fractions (<200 nucleotide length) was used for constructing of cDNA libraries. Differential expressed non-coding RNAs were identified using R package DESeq2.
为探究甲型流感病毒感染过程中小核仁RNA(small nucleolar RNAs, snoRNAs)的表达模式,本研究将人A549细胞以甲型流感A/Puerto Rico/8/1934(H1N1)病毒进行侵染。分别于侵染后24小时与48小时收取样本,采用Illumina NextSeq 500测序平台对侵染细胞开展小RNA测序分析。以同等条件下的模拟侵染(mock-infected)细胞作为对照。提取长度小于200核苷酸的小RNA组分构建cDNA文库,并借助R包DESeq2对差异表达的非编码RNA进行鉴定。
