To investigate how stage-specific activation of canonical Wnt signaling can induce the differentiation of atrioventricular canal cardiomyocytes,We employed single-cell RNA sequencing and ChIP-seq for H3K27ac to interrogate hPSC-derived cardiac cells. Overall design: hPSC-derived cardiac cells at day 11 and day 15 were analyzed using scRNA seq or ChIP-seq.
为探究经典Wnt信号通路(canonical Wnt signaling)的阶段特异性激活如何诱导房室管心肌细胞(atrioventricular canal cardiomyocytes)的分化,本研究采用单细胞RNA测序及针对H3K27ac的染色质免疫共沉淀测序(ChIP-seq),对人类多能干细胞(hPSC)来源的心肌细胞进行了分析。实验整体设计:分别对第11天和第15天的hPSC来源心肌细胞开展单细胞RNA测序或ChIP-seq检测。
