We report the application of RNA-seq technology for high-throughput profiling of gene transcription in BMDMs. By obtaining over six billion bases of sequence from mRNA, we generated genome-wide gene transcription maps of BMDMs infected with recombinant Mycobacterium tuberculosis H37Ra strains. We find that a group of genes is significantly affected by MpbR. This finding indicates that MpbR regulates the host immune. Examination of gene expression level in 3 H37Ra strains infected BMDM cells.

本研究报道了RNA测序(RNA-seq)技术在骨髓源性巨噬细胞(Bone Marrow Derived Macrophages,BMDMs)高通量基因转录谱分析中的应用。通过从信使RNA(mRNA)中获取超过60亿碱基的序列数据,我们构建了感染重组结核分枝杆菌(Mycobacterium tuberculosis)H37Ra菌株的BMDMs的全基因组基因转录图谱。研究发现,一组基因的表达显著受MpbR调控,这一结果提示MpbR可调控宿主免疫。我们对3株感染H37Ra菌株的BMDM细胞的基因表达水平进行了检测。
