Pediatric Medulloblastoma RNA and microRNA sequencing

Medulloblastoma the most common malignant brain tumor in children is divided into four tumor subgroups. The miRNA-mRNA regulatory network in Medulloblastoma is far from being fully understood. Our aims were to investigate miRNA expression regulation in Medulloblastoma subtypes, to assess miRNA-target relationships, and to identify miRNA biomarkers for rapid, specific and cost effective subgroup diagnosis. With these aims, integrated mRNA and miRNA expression analysis was performed on primary tumor samples collected from 9 Medulloblastoma patients (1 - WNT sub-group, 1 - SHH sub-group, 2 - sub-group 3 and 5 - sub-group 4) using small RNA sequencing and RNA-seq.
髓母细胞瘤(Medulloblastoma)是儿童最常见的恶性脑肿瘤,可分为四个肿瘤亚型。目前学界对髓母细胞瘤内的miRNA-mRNA调控网络尚未得到充分解析。本研究旨在探究髓母细胞瘤各亚型的miRNA表达调控规律,评估miRNA与靶基因的相互关系,并筛选可用于快速、特异且经济高效的亚型诊断的miRNA生物标志物。为此,本研究对9例髓母细胞瘤患者的原发性肿瘤样本开展了整合性mRNA与miRNA表达分析,样本涵盖1例WNT亚型、1例SHH亚型、2例第3亚型及5例第4亚型,检测手段包括小RNA测序(small RNA sequencing)与RNA-seq。
