Single cell RNA sequencing of ascending aorta in smooth muscle-targeted elastin deletion

We performed single cell RNA sequencing (scRNA-seq) on ascending aortae from P8 TaglnCre;Elnf/f mice or controls. Overall design: Ascending aortae from mixed background P8 TaglnCre;Elnf/f mice (pooled 4 aortae, mixed sex) and littermate controls (pooled 3 aortae from 1 Elnf/f and 2 Elnf/+, mixed sex) were dissected out. Single cell suspension was prepared by enzymatic digestion and fluorescence-activated cell sorting, and subjected to 10X Chromium 3' v3 library preparation. Sequencing was performed with a Illumina NovaSeq sequencer.
