RNA Sequencing of mouse Purkinje cells across postnatal development

We analyzed Purkinje cell transcriptome dynamics in the developing mouse cerebellum during the first three postnatal weeks, a key developmental period equivalent to the third trimester in human cerebellar development. Our study represents the first detailed analysis of developmental Purkinje cell transcriptomes and provides a valuable dataset for gene network analyses and biological questions on genes implicated in cerebellar and Purkinje cell development. Laser capture microdissection was employed to obtain a highly enriched population of cerebellar Purkinje cells. Deep sequencing was performed on RNA isolated from 1000 Purkinje cells at five developmental timepoints (postnatal days P0, P4, P8, P14 and P21) in triplicate.
本研究针对出生后前三周这一与人类小脑发育妊娠晚期相当的关键阶段,分析了发育中小鼠小脑中的浦肯野细胞(Purkinje cell)转录组动态变化。本研究为首个针对发育阶段浦肯野细胞转录组的系统性详细分析,可为基因网络分析以及围绕小脑与浦肯野细胞发育相关基因的生物学研究问题提供极具价值的数据集。本研究采用激光捕获显微切割(Laser capture microdissection)技术,以获取高度富集的小脑浦肯野细胞群。研究人员针对五个发育时间点(出生后第0、4、8、14及21天)的1000个浦肯野细胞所提取的RNA开展了深度测序,每个时间点设置三次生物学重复。
