Gut microbiome of diabetes

Total DNA of microorganisms was extracted from all feces samples using a QIAamp stool DNA minikit (Qiagen, Germany). The extracted genomic DNA was used to construct an amplicon library by amplifying the V3~V4 region of the 16S rRNA gene. An Illumina Miseq high-throughput sequencing platform (Illumina) was used to sequence the PCR products in the 16S RNA V3~V4 region.

采用德国凯杰（Qiagen）公司的QIAamp粪便DNA迷你试剂盒，从全部粪便样本中提取微生物总脱氧核糖核酸（DNA）。以提取得到的基因组DNA为模板，扩增16S核糖体RNA（rRNA）基因的V3~V4可变区，以此构建扩增子文库。使用Illumina公司的MiSeq高通量测序平台，对16S RNA V3~V4区域的聚合酶链式反应（PCR）扩增产物进行测序。
