Single-cell RNA sequencing of wildtype and miR-146a deficient T cells from mice treated with anti-PD-1 therapy

We observed increased severity of immune related adverse events (irAEs) to anti-PD-1 therapy as well as enhanced T cell effector function in miR-146a deficient mice compared to wildtype (WT) mice. The aim of this experiment was to analyze differences in gene expression between WT and miR-146a deficient T cells at the single cell level. Single-cell RNA sequencing was performed from splenic MACS purified T cells from 2 WT and 2 miR-146a deficient mice that had been treated with anti-PD-1 antibody twice a week for three weeks.To induce irAEs, mice were additionally treated with low-dose LPS once a week.
