Single-Cell Analysis of Dorsal Root Ganglia Reveals Metalloproteinase Signaling in Satellite Glial Cells and Pain

Although satellite glial cells (SGCs) are among the most abundant non-neuronal cells in dorsal root ganglia (DRG), little is known about their heterogenity and functions. We used single cell RNA sequencing (scRNA-seq) to analyze the heterogenity and unique functions of SGCs. DRG tissues from three CD-1 mice were dissected and pooled, single cell suspension was generated by enzymatic and mechanical dissociation, and analyzed using scRNA-seq.
尽管卫星胶质细胞(satellite glial cells, SGCs)是背根神经节(dorsal root ganglia, DRG)中含量最为丰富的非神经元细胞类群之一,但目前学界对其异质性与功能的了解仍十分有限。本研究采用单细胞RNA测序(single cell RNA sequencing, scRNA-seq)技术,对卫星胶质细胞的异质性及独特功能展开分析。实验具体流程如下:解剖并混合3只CD-1小鼠的背根神经节组织,通过酶解与机械解离制备单细胞悬液,随后利用单细胞RNA测序技术完成相关分析。
