Role of MYC acetylated lysine residues in cell transformation and gene-selective regulation [Rat1A]

The functions of MYC acetylation have remained unclear. In this study we identified the major lysine residues of MYC that are acetylated by p300 and GCN5 in cancer cell lines and established the requirment of three specific acetyl-lysine (AcK) residues for MYC transformation of Rat1a fibroblasts and MCF10 mammary epithelial cells. We further uncovered gene-selective regulatory functions and conserved genetic programs and biolofical processes regulated by MYC AcK residues in both cell types.
MYC蛋白乙酰化的功能迄今仍未明确。本研究于癌细胞系中鉴定出了被p300与GCN5催化乙酰化的MYC主要赖氨酸残基,并确立了三个特定乙酰化赖氨酸(Acetyl-lysine, AcK)残基对于MYC介导的Rat1a成纤维细胞与MCF10乳腺上皮细胞转化的必要性。本研究进一步揭示了在两种细胞类型中,由MYC的AcK残基所调控的基因选择性调控功能、保守遗传程序与生物学过程。
