ER, mitochondria and ISR regulation by mt-HSP70 and ATF5 upon procollagen misfolding (P5 parietal bone)

We used single cell RNA sequencing (scRNA-seq) to identify parietal osteoblast cell stress response pathway to misfolding of type I procollagen with Gly610->Cys substitution in the triple helical region of alpha2(I) chain. Parietal bone cells were isolated as described in Gorrell et al, Adv Sci, 2022我们采用单细胞RNA测序（single cell RNA sequencing, scRNA-seq）技术，鉴定了α2(I)链三螺旋结构域中存在甘氨酸610→半胱氨酸替换的I型前胶原错折叠所诱导的顶骨成骨细胞应激反应通路。顶骨细胞的分离方法参照Gorrell等人2022年发表于《Advanced Science》的研究方案。
