Postnatal epigenetic development of mouse intestinal stem cells (RNA-seq)

We used RNA sequencing to quantify the gene expression levels in the intestinal stem cells (ISCs) or their progeny during the suckling period of mouse colon development. Lgr5-EGFP mice were used to identify ISC populations in the colons. RNA sequencing was performed using EGFP labeled Lgr5+ ISCs and epithelial cell adhesion molecule (EpCAM) labeled epithelial cells isolated at the beginning and end of the suckling period (postnatal day 0-P0 and P21).
本研究采用RNA测序(RNA sequencing)技术,定量分析小鼠结肠发育哺乳期内肠道干细胞(intestinal stem cells,ISCs)及其子代细胞的基因表达水平。实验利用Lgr5-EGFP小鼠鉴定结肠中的肠道干细胞群;于哺乳期起始与结束阶段,即出生后第0天(P0)与出生后第21天(P21),分别分离得到经EGFP标记的Lgr5+肠道干细胞以及经上皮细胞黏附分子(epithelial cell adhesion molecule,EpCAM)标记的上皮细胞,并对上述细胞开展RNA测序。
