Transcriptome from Single Nuclei (Mus musculus). Mus musculus strain:Prox1-GFP

A recently developed protocol allows synthesis of double-stranded cDNA from a single cell for use in DNA sequencing. Gene expression is derived from the number of reads mapping to each gene and mutations and mRNA splicing variants are revealed by the sequences. We demonstrate that this method for global transcriptomic analysis can be carried out on the low levels of mRNA present in a single nucleus. Isolated nuclei from a mouse neural progenitor cell line and from dissected hippocampal tissue yielded high quality cDNA that could be used in analysis of gene expression and genotyping. On average, more than 16,000 of the 24,057 mouse protein coding genes were detected from single nuclei.
