制药、化妆品、食品、环境、临床领域发现的微生物经过16S rDNA序列测序后,利用获得的16S rDNA序列通过和本参考数据库序列联配进行细菌种属水平鉴定,以实现污染菌调查、监控、菌种资源的初步筛选,质控菌的确认等目的。微数通过查阅细菌合格发表文献、伯杰氏手册从公共数据库筛选合格发表的物种及代表菌株的16S rDNA序列,并通过进化分析、完整度检查(完整度大于70%)和质量检查(兼并碱基小于2%),建立高质量的16S rDNA数据库,并通过第三方训练的数据库进行比较分、阳性样本序列进行数据库的确认,最终建立高质量16S rDNA数据库,N碱基大于等于10比例低至0.7%以下,小于1100bp区域的不完整序列低至2%以下,优于目前的公开数据库。

Microorganisms isolated from pharmaceutical, cosmetic, food, environmental and clinical fields undergo 16S rDNA sequencing. The acquired 16S rDNA sequences are aligned against this reference database for bacterial identification at the genus and species level, supporting applications including contaminant investigation, monitoring, preliminary screening of microbial strain resources, and confirmation of quality control strains. Weishu developed this high-quality 16S rDNA database by screening 16S rDNA sequences of officially published bacterial species and their representative strains from public databases, with reference to peer-reviewed bacterial literature and Bergey's Manual of Systematic Bacteriology. The screening workflow incorporates phylogenetic analysis, integrity verification (with integrity score >70%) and quality control assessment (with ambiguous bases accounting for <2%). Subsequently, comparative analysis against a third-party trained database and validation using positive control sample sequences were conducted to refine and finalize the database. The finalized high-quality 16S rDNA database exhibits exceptional performance metrics: the proportion of sequences containing ≥10 N bases is as low as <0.7%, the proportion of incomplete sequences with spans shorter than 1100 bp is below 2%, which outperforms all currently available public databases.
