Ethylene signaling mediates host invasion by parasitic plantsParasitic plants form a specialized organ, a haustorium, to invade host tissues and acquire water and nutrients. To understand the molecular mechanism of haustorium development, we performed a forward genetics screening to isolate mutants exhibiting haustorial defects in the model parasitic plant Phtheirospermum japonicum. We isolated two mutants that show prolonged and sometimes aberrant meristematic activity in the haustorium apex, resulting in severe defects on host invasion. Whole genome sequencing revealed that the two mutants respectively have point mutations in homologs of ETHYLENE RECEPTOR 1 (ETR1) and ETHYLENE INSENSITIVE 2 (EIN2), signaling components in response to the gaseous phytohormone ethylene. Application of the ethylene signaling inhibitors also caused similar haustorial defects, indicating that ethylene signaling regulates cell proliferation and differentiation of parasite cells. Larval instar-dependent hemocytes in specialist herbivore Cydalima perspectalis fed on Buxus hyrcana and Buxus microphyllaThe differential interfere contrast (DIC) and light microscopy were used to determine the total (THC) and differential hemocyte count (DHC) in third and sixth instar larvae of Cydalima perspectalis Walker on two boxwood species (Buxus hyrcana and Buxus microphylla). A Neubauer hemocytometer (HBG, Germany) was utilized for THC. The larval hemolymph samples (20 μl) were immediately mixed with 60 μl of anticoagulant solution (0.041 M Citric acid, 0.017 M EDTA, 0.186 M NaCl, 0.098 M NaOH, and pH 4.5) (Amaral et al., 2010). Assays were carried out on larval hemolymph samples individually collected from five larvae (third and sixth instar larvae as replicates) reared on two boxwood species (as treatments). For differential hemocyte counts (DHC), the larvae were submerged in hot distilled water (60°C) for 5 min and then dried with blotting paper. Their hemolymph (20 μl) was collected through a capillary tube from the metathoracic legs of third and sixth instar larvae (< 48 h post ecdysis) fed on two boxwood leaves. According to Gupta (1979), the cell shape, cytoplasmic ratio, cytoplasmic inclusions, and nucleus shape were used for hemocyte identification. The morphometric sizes (length and width) of hemocytes in the third and sixth instar larvae on two boxwood species were determined. In addition, data from total hemocyte (THC) and different hemocyte count (DHC) of two different larval instars fed on the same boxwood species were analyzed using t-test at p < 0.05. Furthermore, total (THC) and different hemocyte counts (DHC) of the same larval instars fed on two boxwood species (B. hyrcana and B. microphylla) were also compared using t-test.
Reduced Germination of Orobanche cumana Seeds in the Presence of Arbuscular Mycorrhizal Fungi or Their ExudatesBroomrapes (Orobanche and Phelipanche spp) are parasitic plants responsible for important crop losses, and efficient procedures to control these pests are scarce. Biological control is one of the possible strategies to tackle these pests. Arbuscular Mycorrhizal (AM) fungi are widespread soil microorganisms that live symbiotically with the roots of most plant species, and they have already been tested on sorghum for their ability to reduce infestation by witchweeds, another kind of parasitic plants. In this work AM fungi were evaluated as potential biocontrol agents against Orobanche cumana, a broomrape species that specifically attacks sunflower. When inoculated simultaneously with O. cumana seeds, AM fungi could offer a moderate level of protection against the broomrape. Interestingly, this protection did not only rely on a reduced production of parasitic seed germination stimulants, as was proposed in previous studies. Rather, mycorrhizal root exudates had a negative impact on the germination of O. cumana induced by germination stimulants. A similar effect could be obtained with AM spore exudates, establishing the fungal origin of at least part of the active compounds. Together, our results demonstrate that AM fungi themselves can lead to a reduced rate of parasitic seed germination, in addition to possible effects mediated by the mycorrhizal plant. Combined with the other benefits of AM symbiosis, these effects make AM fungi an attractive option for biological control of O. cumana.

Cuscuta pentagona (dodder) is a parasitic plant that forms symplastic connections to its host and takes up host mRNAs. We sequenced transcriptomes of Cuscuta growing in association with genetically distinct hosts, Arabidopsis and tomato, to characterize mRNA transfer between species and found that mRNAs move in high numbers and in a bidirectional manner.
Time course of trans-species miRNAs from Cuscuta campestris grown on Arabidopsis thaliana. Time course of trans-species miRNAs from Cuscuta campestris grown on Arabidopsis thalianaCuscuta campestris is an obligate stem parasite which uses an organ called the haustoria to divert water and photosynthates from the host. Previously, we have identified that at the haustorial interface between Cuscuta campestris and Arabidopsis thaliana, miRNAs generated by the parasite are able to move into the host and regulate host gene expression. This study identifies how long after attachment does trans-species miRNA transcription begin in Arabidopsis thaliana and Solanum lycopersicum, as well as identifying which stage of haustoria development they become detectable. A time course was performed by harvesting interfaces every 24 hours post attachment, and samples were subjected to RNA extraction and sRNA sequencing. We have identified that in Arabidopsis thaliana, trans-species miRNAs become detectable two days post attachment. In S. lycopersicum, some trans-species miRNAs are detectable one day post attachment, but all become detectable by day 2. Secondary siRNA accumulation was detected four days post attachment in both hosts. In order to determine which stage of haustoria development trans-species miRNAs become detectable, vibratome sectioning was performed on the haustorial interfaces of both hosts. By looking at the morphology of the developing haustoria, it was determined that trans-species miRNAs become detectable during the adhesive phase. This suggests that trans-species miRNA production is one of the first steps of haustoria development, as the parasite tissue has not started to invade host tissue before they become detectable. Overall design: Samples were taken every 24 hours for 15 days (days 0 through 14) after coiling of the parasite around the host stem. There were between 1 and 3 replicate samples per time point.
Dodder transmits salt-induced systemic signals between cucumber hostsCuscuta spp. (dodders) are shoot parasites that connected to vascular tissue of hosts extracting water, nutrients, metabolites, protein, mRNAs, evens virus from hosts. Table_5_Phenotypic and histological analyses on the resistance of melon to Phelipanche aegyptiaca.docxMelon (Cucumis melo L.) is an economically important crop in Xinjiang, China, but its production is constrained by the parasitic plant Phelipanche aegyptiaca that attaches to the roots of many crops and causes severe stunting and loss of yield. Rhizotron, pot, and field experiments were employed to evaluate the resistance of 27 melon cultivars to P. aegyptiaca. Then, the resistant and susceptible cultivars were inoculated with P. aegyptiaca from six populations to assess their resistance stability and broad spectrum. Further microscopic and histological analyses were used to clarify the resistance phenotypes and histological structure. The results showed that Huangpi 9818 and KR1326 were more resistant to P. aegyptiaca compared to other cultivars in the rhizotron, pot, and field experiments. In addition, compared to the susceptible cultivar K1076, Huangpi 9818 and KR1326 showed broad-spectrum resistance to six P. aegyptiaca populations. These two resistant cultivars had lower P. aegyptiaca biomass and fewer and smaller P. aegyptiaca attachments on their roots compared to susceptible cultivar K1076. KR1326 (resistant) and K1076 (susceptible) were selected to further study resistance phenotypes and mechanisms. Germination-inducing activity of root exudates and microscopic analysis showed that the resistance in KR1326 was not related to low induction of P. aegyptiaca germination. The tubercles of parasite on KR1326 were observed slightly brown at 14 days after inoculation (DAI), the necrosis and arrest of parasite development occurred at 23 DAI. Histological analysis of necrosis tubercles showed that the endophyte of parasite had reached host central cylinder, connected with host xylem, and accumulation of secretions and callose were detected in neighbouring cells. We concluded that KR1326 is an important melon cultivar for P. aegyptiaca resistance that could be used to expand the genetic basis of cultivated muskmelon for resistance to the parasite.

How do host-plant use and seasonal life cycle relate to insect body size: A case study on European geometrid moths (Lepidoptera: Geometridae)We used European geometrid moths (> 630 species) as a model group to investigate how life history traits linked to larval host plant use (i.e., diet breadth and host-plant growth form) and seasonal life cycle (i.e., voltinism, overwintering stage, and caterpillar phenology) are related to adult body size in holometabolous insect herbivores. To do so, we applied phylogenetic comparative methods to account for shared evolutionary history among herbivore species. We further categorised larval diet breadth based on the phylogenetic structure of utilised host plant genera. Our results indicate that species associated with woody plants are, on average, larger than herb feeders and increase in size with increasing diet breadth. Obligatorily univoltine species are larger than multivoltine species, and attain larger sizes when their larvae are restricted to the early season. A long-distance inhibitory system regulates haustoria numbers in parasitic plantsTranscriptomic response changes following infection in shoots of the parasitic plant Phtheirospermum japonicum. Transcriptomic changes in the shoots of 17 days old Phtheirospermum japonicum that has been infecting 7 days old Arabidopsis thaliana (Col-0) for 10 days in vitro.
