Twenty-two BAC plasmids carrying sequences from chromosome 2 of the silkworm were used to assemble a 2.77 Mb linear plasmid in yeast. Both the 22 original BAC plasmids and the 5 assembled plasmids were sequenced to validate the assembly method.
