For MutMap, leaves of 50 mutant-phenotype plants from the BC1F2 population were pooled in equal proportions, forming a mutant bulk for whole-genome resequencing. The wild-type II-32B was also sequenced as a control. For WGBS, stigmas of both mutant her1 and wild-type II-32B were collected, genomic DNA was extracted, subjected to sodium bisulfite conversion, and sequenced to obtain whole-genome methylation profiles.
