Genetic mapping of the zebrafish b1115 mutant was done on a cross of two b1115 carriers. Phenotypic mutants were identified at 5 days post-fertilization and 5 individuals were pooled together prior to DNA extraction. SbfI RAD-tag libraries were prepared from the cross parents, 9 wild type pools, and 34 b1115 mutant pools and sequenced on an Illumina HiSeq 2000 to generate 100-nucleotide single-end reads for bulk segregant analysis.
