In order to accurately design CRISPR gRNAs, HDR vectors and other molecular reagents to more accurarately target common parental lines used by the Gene Editing team (i.e. KOLF2-C1 ARID2 corrected clone D5 and HAP1 LIG4-/- cell lines) a sequenced verification is required. In order to accurately design CRISPR gRNAs, HDR vectors and other molecular reagents to more accurarately target common parental lines used in gene editing we have whole genome sequenced a derivative of the human iPSC line (KOLF2-C1) and the commonly used HAP1 (haploid) cell lines. Haplotype data from the iPSC line will provide information on genotype and allele specidifc polymoriphisms and mutations that will be important when deisgning gene editing targeting strategies and indeed their potentila for downstream experiements such as differentiation. HAP1 lines are evidenced as being unstable during long term culture. We have whole genome sequenced our HAP1 LIG4-/- to rapidly facilitate the generation of molecular regaents for gene targeting.
