We analyzed transcriptome of distinct SF3B1 hotspot mutations in myelodysplastic neoplasms (MDS). RNA sequencing was performed on CD34+ bone marrow cells from 88 MDS patients (56 with SF3B1 mutations, 32 wild-type) and 13 healthy donors. Additionally, CRISPR-engineered NALM-6 cell lines harboring individual SF3B1 hotspot variants (wild-type, K700E, K666N, or H662Q) were sequenced in triplicates.
