Activation of cGAS-STING signaling in cancer cells requires abundant cytosolic DNA that is induced through cell intrinsic or treatment associated DNA damage. However, clinical trials based on this rationale to increase immune infiltration and benefit immunotherapy is challenging, highlighting gaps in understanding the link between DNA damage and immunotherapy. Here, we identify ubiquitination-directed cytosolic DNA degradation as a critical determinant for cGAS-STING activation following DNA damage. This Subseries contains RNA sequencing data from the human prostate cancer cell line PC-3, designed to systematically profile transcriptomic changes resulting from inhibition of multiple deubiquitinase (DUB) enzymes. These data aim to uncover DUB-mediated regulatory mechanisms that modulate cytosolic DNA stability and cGAS-STING signaling, with the goal of identifying novel molecular targets for combination immunotherapy in prostate cancer.
