Immune thrombocytopenia (ITP) is characterized by overproduction of anti-platelet autoantibodies through elusive mechanisms. Here, we investigated how central B-cell tolerance mechanisms affect autoantibody production in ITP. Our study employed integrative single-cell RNA and BCR sequencing (scRNA/BCR-seq) to systematically characterize the immunoglobulin (IG) repertoire across different developmental stages in bone marrow B cells. Notably, we discovered reduced receptor editing within IG kappa (IGK) genes, a critical process ensuring B cell tolerance.
