Virus-like particles (VLPs) have been engineered to package Cas9 protein or pre-assembled Cas9-guide RNA ribonucleoprotein complexes for the purposes of genome editing in mammalian cells. Lentivirus packaging Cas9 (Cas9-VLPs) were produced and use to treat the indicated cell lines. DNA was isolated 3 days post treatment, and targeted genomic regions were PCR amplified, and subjected to deep sequencing (Illumina MiSeq).
