Expression profiling of Dicer loss-of-function macrophages

The role of microRNAs in the inflammatory response of macrophages to LPS was adressed by expression analysis in DicerKO (LysMcre/+;Dicerfl/fl;ApoE-/-) vs wildtype (LysMcre/+;Dicer+/+ApoE-/-) macrophages when challenged either with vehicle (PBS) or LPS. Overall design: Bone marrow-derived macrophages of male LysMcre/+;Dicerfl/fl;ApoE-/- and there litter mate control mice (LysMcre/+;Dicer+/+;ApoE-/-) were isolated and differentiated in vitro. After 6h treatment with either vehicle (PBS) or 100 ng/ul LPS, macrophages were harvested. The RNA was isolated and sequenced. 3 replicates per genotype and treatment were analyzed.
