Infiltrating T cells express FOXP3 and IL10.

(A) Western Blot analysis of CD3+ cells retrieved from skeletal muscles at different time points after muscle injury (representative blots of two experiments are shown). Muscle T cells express FOXP3 at all time points analyzed. (B) T cells isolated from skeletal muscle after CTX injury express high level of Il10. CD3+ cells from muscle did not express Il2, Il4 or Il23 and showed variable expression of Tgfβ, Il6, Il27, Ifnγ and Il17 (n = 2 independent experiments, mean ± SEM). (C) Flow cytometric analysis of FOXP3 expression on CD3+ T cells. Representative plots of each time point analyzed are shown. The percentage of CD3+FOXP3+ cells in the injured muscle were quantified and compared to untreated spleens (n ≥ 3 mice per time point, mean ± SEM; t-test) (NT = CD3+ cells from untreated mouse spleen).
