The CRISPR/Cas9 is a new system that can introduce site-specific double strand breaks. In this system, sequence specificity is achieved by 20-nucleotide RNA molecules that guild endonuclease to the specific loci. This method is considerably easier to generate a new construct targeting a new locus than existing technologies such as ZFNs and TALENs and has a potential to generate genome-wide guild RNA libraries. However, because the sequence specificity is determined by only 20 bp, off-target cleavage by this system is generally concerned. In this study, we will analyse likelihood of off-target cleavages by the CRISPR/Cas9 system on computationally predicted off-target sites.
