A high-throughput dilution-to-extinction culturing method (HTC) was applied to eight freshwater samples to isolate diverse freshwater heterotrophic bacteria. The current study applied amplicon pyrosequencing targeting V1-V3 region of the 16S rRNA gene to investigate microbial diversity and compare it with results obtained from bacterial sequences isolated by HTC.
