Approximately 1500, 3000 and 7700 sorted B cells were processed on a 10x Chromium Controller device using Next GEM Single Cell V(D)J v1.1 Reagent Kits (10x Genomics, Pleasanton, CA, USA) following the manufacturer's instructions. GEM were processed, cDNA was generated and antibodies heavy and light chains were amplified using cattle-specific constant region primers paired with Read1 primer.
