This paper studies the impact of synonymous codons on gene expression variation. By constructing a GFP reporter gene library and expressing it in Escherichia coli, the Sort-seq method was used to analyze the GFP coding sequences. The results show that codon usage significantly affects the average expression level and variation of GFP, but has little impact on intercellular variation of high-expression proteins, indicating that codon optimization can be carried out without affecting gene expression noise, which is of great significance for biotechnological applications.
