To evaluate the specificity of DropCRISPRa system in gene activation, we co-transfected with IL1RN-targeted gRNA and DropCRISPRa (FUSIDR-VP64), and carried out whole-transcriptome RNA-seq of HEK293T cells. We also transfected with DropCRISPRa without IDR and TF as negative control for each case. The library preparation and next-generation sequencing were performed by GENEWIZ.
