Type 2 Diabetes (T2DM) is a chronic metabolic disorder characterized by hyperglycemia and peripheral insulin resistance. Further, insulin resistance in T2DM leads to increased hepatic glucose production by the process of gluconeogenesis. PCK-1 is the key limiting enzyme of gluconeogenesis and hence, functionalized graphene sheet based delivery of PCK-1 siRNA was done intravenously in T2DM mice. Liver tissue from control, T2DM and Therapeutic (TG) mice were then used for RNA-sequencing to study the cross talk of PCK-1 silencing on different hepatic pathways. To the best of our knowledge, no study has yet been done to study the the hepatic transcriptome profile of PCK-1 silenced mice.
