Total Gene expression analysis of H3f3b constitutive knockout testis RNA

Total gene expression analysis was performed on RNA from testes extracted from two litters of constitutive homozygous and heterozygous H3f3b knockout mice compared to WT littermates. Constitutive knockout mice were obtained by mating H3f3b Fl/Fl mice to Zp3Cre mice. Heterozygous knockout mice were crossed until homozygous knockout mice were obtained. Testes were surgically removed from 8 week old homozygous knockouts, heterozygous knockouts and WT, and RNA was extracted from one testis from each mouse for total gene expression.
本研究对两窝组成型敲除(constitutive knockout)纯合、杂合H3f3b基因敲除(knockout, KO)小鼠的睾丸提取RNA开展全基因表达分析,实验对照组为同窝野生型(Wild Type, WT)小鼠。 组成型敲除小鼠通过将H3f3b Fl/Fl小鼠与Zp3Cre小鼠交配构建获得。研究人员对杂合敲除小鼠进行多代繁育,直至获得纯合敲除小鼠。分别对8周龄的纯合敲除、杂合敲除小鼠及野生型小鼠实施外科睾丸摘除术,每只小鼠取单侧睾丸提取RNA,用于全基因表达检测。
