In these experiments we transfected HEK 293T cells with CFTR Y122X and various combinations of guide RNAs and site-directed RNA editing enzymes in order to correct the mutation. RNA was extracted from each condition and sequenced using the Illumina platform. DNA was also isolated and sequenced from a separate plate from the same cells.
