采集稻田土-水界面的周丛生物膜到实验室中进行不同磷素浓度的室内模拟培养。分别设置高、中、低三个磷素浓度，并且磷素浓度为唯一变量及限制因子。细胞在人工气候室中进行25℃，12小时光照/天的培养。每个处理设置4个重复。培养箱长30cm宽20cm。于细胞生长对数期采集细胞培养液样品。主要步骤如下：轻微摇晃培养箱，用注射器抽取100ml培养液到离心管中，经过离心，过0.22微米滤膜后，获得周丛生物膜的胞外
